Rapamycin and aging: what human trials tell us

Rapamycin (sirolimus) has mixed human evidence for healthy aging. PEARL found no improvement in its main body-fat measure over 48 weeks, and RAPA-EX-01 found no added functional benefit alongside exercise. Everolimus improved selected vaccine responses, while a larger RTB101 trial missed its respiratory-illness endpoint. Healthy Longevity Clinic weighs these results by function, infection prevention and tolerability.

Metformin for healthy aging: what the evidence supports

Metformin helps control type 2 diabetes and reduced new diabetes cases by 31% relative to placebo in the Diabetes Prevention Program. That is a meaningful healthspan benefit for people at high risk. This review distinguishes established prevention from newer frailty findings, exercise trade-offs and the proposed TAME trial, which aims to test broader prevention of age-related disease.

GLP-1 medicines: benefits beyond weight loss

Semaglutide has shown benefits beyond weight loss. In SELECT, serious cardiovascular events occurred in 6.5% of treated adults versus 8.0% with placebo; participants had cardiovascular disease and overweight or obesity, without diabetes. Kidney and symptom benefits in other defined groups add to the healthspan picture. Healthy Longevity Clinic examines who benefited, how much, and which risks affect the decision.

Retatrutide: what the weight-loss trials show

Retatrutide has produced substantial weight loss in clinical trials. In one phase 3 study, Lilly reported an average reduction of 28.7% versus 2.1% with placebo over 68 weeks in an analysis assuming continued treatment. Published research also shows improved glucose control in type 2 diabetes. These benefits warrant attention, but cardiovascular protection, long-term safety and an approval date remain unsettled.

Senolytics in humans: what the trials show

Dasatinib–quercetin, fisetin and the experimental drugs UBX0101 and UBX1325 test whether removing senescent cells can improve bone, lung, joint or eye health. Results are mixed: several trials missed their main goals, while narrower biological and visual signals warrant follow-up. Healthy Longevity Clinic compares the prospects for fewer fractures, better mobility and preserved vision, together with treatment risks.

Longevity drugs: what human trials show

Metformin, semaglutide and dapagliflozin reduce diabetes or serious heart and kidney events in defined patient groups. Evidence for rapamycin (sirolimus), the senolytic combination dasatinib–quercetin and the NAD precursor nicotinamide riboside (NR) remains early or mixed. This September 2026 overview also compares testosterone and menopausal hormone therapy. Healthy Longevity Clinic assesses their healthspan value through disease prevention, physical function and symptom relief.
